Keratin intracellular concentration revisited: Implications for keratin function in surface epithelia

چکیده
In 1978 Sun and Green carried out a densitometry-based analysis of polyacrylamide gelelectrophoresed samples and reported that keratin proteins account for 25~35% of total cellular proteins in human epidermal keratinocytes serially passaged in primary culture. This represents an astounding figure with very significant implications for the structural support role of keratin intermediate filaments (IFs) in epidermis, and its regulation by posttranslational modifications and/or interaction with associated proteins. We report here on an effort to investigate this issue further, using a distinct method of keratin quantitation in sorted but otherwise native cell populations, and taking advantage of the significant progress made in our understanding of the structure of keratin filaments and differential regulation of keratin expression in epidermis.

Keratin 13 is a more specific marker of conjunctival epithelium than keratin 19
Introduction To evaluate the expression patterns of cytokeratin (K) 12, 13, and 19 in normal epithelium of the human ocular surface to determine whether K13 could be used as a marker for conjunctival epithelium. Methods: Total RNA was isolated from the human conjunctiva and central cornea. Those transcripts that had threefolds or higher expression levels in the conjunctiva than the cornea wer...
متن کاملPairwise assembly determines the intrinsic potential for self-organization and mechanical properties of keratin filaments.
Most type I and II keratin genes are spatially and temporally regulated in a pairwise manner in epithelial tissues, where they represent the major structural proteins. Epithelia can be partitioned into simple (single-layered) and complex (multilayered) types. We compared the structural and mechanical properties of natural keratin polymers occurring in complex (K5-K14) and simple (K8-K18) epithe...
متن کاملOral cytokeratins in health and disease.
The dynamics of oral mucosa is known by its inherent defensive nature. Certain areas demand tough shield when subjected to mechanical insults. This is met by structural scaffolding material referred as cytoskeleton comprised of intracellular protein filaments called cytokeratins in the surface squames of oral epithelia. They also equally contribute towards the architecture of odontogenic appara...

Keratin intermediate filament proteins form cytoskeletal scaffolds in epithelia, the disruption of which affects cytoarchitecture, cell growth, survival, and organelle transport. However, owing to redundancy, the global function of keratins has not been defined in full. Using a targeted gene deletion strategy, we generated transgenic mice lacking the entire keratin multiprotein family. In this ...
